Sclareol and ambroxide: the molecules behind ambroxan

Ambroxan is a scent molecule modelled on ambergris. Ambergris forms in the digestive tract of the sperm whale; its scent comes from breakdown products of the odourless ambrein, created by air, seawater and sunlight. The most important of these is ambroxide, with the formula C₁₆H₂₈O. Around 1950, chemists at the Swiss fragrance manufacturer Firmenich determined its structure and filed a patent for a synthesis. The starting material is still mostly sclareol from clary sage (Salvia sclarea). It is oxidised to the lactone sclareolide, reduced to a diol and finally closed into a ring ether. As a raw material, the molecule is known under names such as Ambrox® or Ambroxan®.

In a fragrance, ambroxan is warm, clean and ambery, with woody, slightly musky and almost salty, mineral facets. It is exceptionally long-lasting and gives a composition volume and structure. With cedarwood or vetiver it feels dry and clear, with labdanum or benzoin warm and ambery, with musk soft and close to the skin.

In this selection, ambroxan sits in the fragrance pyramid – mostly in the base note.

How the note behaves

Woody & mineral

Ambroxan with cedarwood, vetiver or cashmere wood. Dry, clear and modern.

Warm & ambery

Ambroxan with labdanum, benzoin or vanilla. Rounded, soft and radiantly warm.

Fresh & clean

Ambroxan with bergamot, grapefruit or musk. Bright, airy and remarkably enduring.

Curious factOnly about one in a hundred sperm whales produces ambergris at all – the natural model for ambroxan.

Knowledge to share

Myth or truth?

1 / 3

Statement 1

Ambroxan is obtained from whales.

MYTH

Myth. Ambroxan is manufactured – mostly from sclareol from clary sage. The sperm whale’s ambergris serves only as the olfactory model.

Statement 2

The ambroxan molecule also occurs in natural ambergris.

TRUTH

True. It forms there when the odourless ambrein is broken down by air, seawater and light, and is regarded as the key odorant of ambergris.

Statement 3

Ambroxan fades quickly like a top note.

MYTH

Myth. Ambroxan is highly tenacious and often remains perceptible on a smelling strip for weeks; it belongs in the base note.

Frequently asked questions

Is ambroxan natural or synthetic?

Ambroxan is manufactured, mostly from sclareol, a constituent of clary sage. The substance itself also occurs in natural ambergris, but for perfume it is not obtained from it. Today there are also biotechnological production routes.

What does ambroxan smell like?

Warm, clean and ambery, with woody and slightly musky tones. Many also describe a dry, almost salty, mineral note. The molecule is effective even at low dosages.

How long does ambroxan last on the skin?

Ambroxan is one of the most tenacious fragrance materials and acts as a fixative in the base. On a smelling strip it often remains perceptible for weeks. The best test is on your own skin: with every bottle of 30 ml or more we include a 2 ml sample of the same fragrance.
